English: Mechanism of action of asciminib. (A) Under normal conditions, ABL1 activity is autoregulated by binding of myristoylated N-terminus to the myristoyl pocket of the kinase domain. (B) In CML, ABL1 kinase is constitutively activated due to loss of regulatory function with BCR::ABL1 fusion oncoprotein formation (left side). Asciminib binds to the myristoyl pocket of the ABL1 kinase domain, inducing an inactive conformational change and inhibiting kinase activity. |
